Antibacterial Effect of Chitosan and Grape Seeds Extracts against Staphylococcus aureus and Escherichia coli

Abstract
The antimicrobial activity of chitosan, alcohol and grape seeds extracts against Staphylococcus aureus and Escherichia coli was studied to investigate the use of chitosan and grape seeds extracts as natural resources for sanitizing and extending shelf life of food. The antimicrobial activity of acetic acid and citric acid did not show significant difference against test microorganims, respectively. Chitosan N-3 (deacetylated degree: 98.8%) showed the strongest antimicrobial activity against test microorganisms among 6 kinds of tested chitosans. Antimicrobial activity of grape seeds extract was better than that of alcohol and chitosan. The mixture (40% alcohol containing 0.1% chitosan) inhibited about 4 log cycle of test microorganisms and viable cell number reduced about 99.999% after treatment for 5 minutes at 20oC, respectively. The viable cell of S. aureus and E. coli did not detect after treatment for 5 minute in the mixture(40% alcohol containing 0.1% chitosan) with grape seeds extract regardless its contration(100 ppm and 200 ppm), respectively. The optimum mixing ratio of antimicrobial agent to use as sanitizer and ingrents for sanitizing and extending shelf life of food was 0.1% chitosan, 40% alcohol and 100 ppm grape seeds extract.
